You need to increase your total winnings at the 3 main casinos on the strip to the following amounts -
Gommorah - 9000 chips
The Tops - 10000 chips
Ultra-Luxe - 15000 chips
Your gambling will be interrupted by the casino staff several times as you increase your winnings, and eventually you will be banned from gambling further (NOTE: you can still use everything else at the casino once banned, you just can't gamble)
Increasing your Luck stat helps a lot with this (there is an implant at the New Vegas Medical Clinic if you don't want to invest perks in Intensive Training, and there are a few clothing items in the game that boost Luck).
Blackjack is quite quick to do this with. The max bet is 200 chips but you can double down to earn 400 on a win (which is very effective with high Luck). My strategy was to stay on 19+, and double down on just about anything else.
