This achievement is straight forward and easy to get since the update that unlocks all of the Heroes for everyone to play.
NOTE: the achievement says play all

BUT you are required to play all the launch (When the game first come out) villains too.
The best and easiest way to get this achievement fast is by going into a Heroes vs Villains game and every time you die switch character. Make sure you stay in the lobby after the game ends so you switch sides so you can play the heroes or villains that you're missing!
