This is for using one type of gear (short sword, scythe, sniper gun, tower shield, whatever) for a total of 500 missions. If you haven't unlocked this by the end of the game, play the Challenge Mission called "Today's Fortune"...the mission auto-completes after 1 second, and counts. I had about 70 missions left so this made it really quick!
